    摘要:

    以小粒野生稻核DNA为材料构建了一个可转化人工染色体文库,获得了5000个克隆,平均插入片段约45 kb。稳定性检测结果表明,小粒野生稻基因组DNA能够在TAC载体中稳定存在。基于已克隆抗性基因的保守序列设计探针,利用菌落杂交的方法,对文库进行筛选。结果表明,该文库可以用于抗性基因的筛选。

    Abstract:

    A transformation-competent artificial chromosome (TAC) library was constructed from the genomic DNA of orazy.mimuta. This library is composed of 5,000 clones while the average insertion fragment size is 45 kb. Stability test indicated that genome DNA of Orazy.mimuta was stably existence in the pYLTAC27 carrier. A probe was designed according to the sequences of cloned resistance gene. With the colony hybridization method, the genome library was screened. The result shows that resistance gene can be screened in this library.
